چکیده

Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 29 Prunus spp. rootstocks. The DNA from the rootstocks was analyzed using five pre-selected SSR primers (UDP96-005, UDP96-008, UDP96013, UDP96-18 and UDP98-414) and revealed 81 alleles, which allowed each genotype to be identified. The UDP96-005 marker generated the most information, i.e., 23 well-distributed, polymorphic alleles among all genotypes. The 21 polymorphisms produced by UDP96-013 occurred mainly as a result of high degree of variability among genotypes of the Prunophora subgenus. In the dendrogram, the five markers allowed the 29 rootstocks to be grouped into subgroups corresponding to the subgenus they belong to, either Prunophora or Amygdalus. Suitable cophenetic correlation coefficient (r=0.82) and good bootstrapping fitting value among the Prunophora subgroup cultivars were obtained. SSR markers proved to be efficient and reliable for the molecular characterization of Prunus spp. rootostocks.

First Report of a set of Genetic Identities in Prunus Rootstocks by SSR Markers

Prunus rootstocks play an important role in modern horticulture and commercial orchards owing to their responsibility for a wide range of characters from compatibility with cultivars to adaptation to biotic and abiotic stresses. In this study, Thirty Prunus rootstock samples were tested by 25 microsatellite markers in order to identify the genetic identity and relationships among them.17 SSR ma...

Application of Random Amplified Microsatellite Polymorphism (RAMP) in Prunus Characterization and Mapping

Random amplified microsatellite polymorphism (RAMP) is a PCR-based marker which uses a combination of two classes of markers: Simple sequence repeat (SSR) and Random amplified DNA polymorphism (RAPD) markers. RAMP has been demonstrated to be a potentially valuable molecular marker for the study of genetic relationships in cultivated plant species. The objective of this study was to optimize the...

Assessment of the Genetic Diversity of Almond (Prunus dulcis) Using Microsatellite Markers and Morphological Traits

The genetic diversity among 56 almond (Prunus dulcis) genotypes was analysed using 35 microsatellite markers and 14 morphological traits. Analysis of morphological traits revealed a wide range of variation among the studied genotypes. Out of 35 simple sequence repeats (SSRs) markers, 25 were polymorphic, producing 215 alleles that varied from 2 to 16 with an average of 8.76 alleles per locus. R...

Host Suitability of Eight Prunus spp. and One Pyrus communis Rootstocks to Pratylenchus vulnus, P. neglectus, and P. thornei.

The effects of Pratylenchus vulnus on rootstocks of eight commonly used Prunus spp. and one Pyrus communis were evaluated under greenhouse conditions during a 15-month period. In a first experiment, two almonds (Moncayo and Garrigues), one peach (GF-305), and two peach-almond hybrids (GF-677 and Adafuel) inoculated with 2,000 nematodes per plant proved to be good hosts of P. vulnus. Highest (P ...

Peach Rootstocks for the United States: Are Foreign Rootstocks the Answer?

New foreign rootstocks for peaches [Prunus persica (L.) Batsch] are now being introduced into the United States through commercial nurseries for future sales to stone fruit growers. Almost all of these rootstocks are complex Prunus L. hybrids that are propagated vegetatively. Past experience with foreign Prunus rootstocks has shown that extensive testing is critical to avoid potential problems ...
